Real-Time Monitoring of AAV Empty: Full Capsid Separations using Near Infrared High-Precision Tunable Laser Spectroscopy

Adeno-associated virus (AAV) vectors are a cornerstone of gene therapy, but their efficacy and safety depend heavily on the ratio of empty-to-full capsids. Traditional analytical methods for monitoring this critical quality attribute (CQA) during purification processes, such as ultracentrifugation and mass photometry, can be time-intensive and laborious, while rapid methods such as AEX-HPLC and SEC-MALS can typically not resolve partial from full capsids. Here, we present a novel approach utilizing Near Infrared (NIR) High-Precision Tunable Laser Spectroscopy (HPTLS) for real-time monitoring of AAV empty-to-full capsid ratios using chromatography. Nirrin Technologies’ proprietary HPTLS technology enables high-resolution spectral separation of capsid species, providing quantification of full and empty capsids in real-time. This method eliminates the need for extensive sample preparation while maintaining accuracy comparable to traditional methods. The integration of HPTLS into downstream bioprocessing workflows enhances process control, enabling rapid decision-making and reducing production timelines. This work demonstrates the potential of HPTLS to transform AAV purification through non-invasive, real-time analytics.
